    Neurocognitive Modulations of Lexical Access during Speech Production in Social and Semantic Context

    Der Sprechakt teilt sich in zwei Vorgänge: Zunächst muss das richtige Wort aus dem mentalen Lexikon abgerufen werden und anschließend wird es in der Kommunikation verwendet. Zur Erforschung des ersteren Vorgangs werden oft Ein-Personen-Studien verwendet, in denen durch Beobachten der Reaktion auf Stimuli (z. B. Bilder) die Mikrostruktur des lexikalisch- semantischen Systems beleuchtet wird. Für die Anwendung von Sprache in der Kommunikation hingegen nutzt man Partnerexperimente, um die Koordination zwischen den Gesprächspartnern zu beobachten und zu ergründen, wie sich gegenseitiges Verstehen und biografisches Wissen darauf auswirken. Wenig erforscht ist aber, wie ein von einem Gesprächspartner eingebrachter Bedeutungskontext die traditionell in Ein-Personen-Studien untersuchten lexikalisch-semantischen Effekte beeinflusst. Im Rahmen meiner Dissertation möchte ich die Lücke zwischen den beiden Forschungsansätzen schließen, indem ich einen kommunikativen Kontext in etablierte Paradigmen der Bildbenennung integriere. Hierzu betrachte ich zunächst klassische semantische Kontexteffekte, die durch nähere oder entferntere kategorische Relationen zwischen Begriffen hervorgerufen werden (Studie 1), um anschließend lose thematische Beziehungen zu untersuchen, die mit alltäglichen Ereignissen verbunden sind (Studie 2 & Studie 3). Um die hochgradig verflochtenen Ebenen der lexikalischen und semantischen Verarbeitung voneinander zu trennen, habe ich ereigniskorrelierte Hirnpotentiale (ERPs) eingesetzt, um die elektrophysiologischen Signaturen des konzeptuellen Primings und der lexikalischen Auswahl zu verfolgen. Die vorliegende Arbeit liefert sowohl theoretische als auch praktische Beiträge. Erstens stützen unsere Ergebnisse die theoretischen Annahmen, dass sich semantisches Priming und lexikalische Interferenz vorübergehend überschneiden und gemeinsam das Benennungsverhalten in einem Trade-off beeinflussen. Auch die Gegenwart eines Kommunikationspartners kann Auswirkungen auf dieses Zusammenspiel haben. Zweitens ergänzen diese Ergebnisse die aktuelle Literatur zu verschiedenen Arten von semantischen Beziehungen, wie z. B. Nulleffekte für entfernte Beziehungen und Kontexteffekte, die systematisch mit der Stärke der Verwandtschaft zunehmen. Und schließlich bietet unser neuartiges Design eines kommunikativen Kontextes ein praktisches Instrument, um die Lücke zwischen Ein-Personen-Studien und Kommunikationsstudien zu schließen. Alles in allem tragen diese Ergebnisse zu einem besseren Verständnis der neuronalen Mechanismen unseres Sprachproduktionssystems bei, das in der Lage ist, sich flexibel sowohl an sprachliche als auch an soziale Kontexte anzupassen.Speaking could be divided into two processes: first, the correct word must be retrieved from the mental lexicon, and then it is used in communication. To study the former process, single-person studies are often used, in which the microstructure of the lexical-semantic system is illuminated by observing reaction times to name stimuli (e.g., pictures). For the language use in communication, on the other hand, partner experiments are used to observe coordination between interlocutors and to explore how mutual understanding and biographical knowledge affect it. However, how a meaningful context brought by an interlocutor influences the established lexical-semantic effects from single-person studies remains underexplored. Within the scope of my dissertation, I aim to bridge the gap between these two research approaches by integrating a communicative context into well-established picture naming paradigms. To this end, I first investigate classic semantic context effects induced by close or distant categorical relations (Study 1), and then examine loose thematic relations associated with everyday events (Study 2 & Study 3). To separate the highly intertwined strata of lexical and semantic processing, I used event-related brain potentials (ERPs) to track the electrophysiological signatures of conceptual priming and lexical selection. The present work makes both theoretical and practical contributions. First, our results support the theoretical assumptions that semantic priming and lexical interference temporarily overlap, and jointly modulate naming behavior in a trade-off. Such interplay may be greatly influenced by the presence of a communicating partner. Second, these findings add to the current literature on different types of semantic relations, such as null effects for distant relations and context effects that systematically increase with the strength of relatedness. Finally, our novel design of a communicative context provides a practical tool to bridge the gap between single-person studies and communication studies. All in all, these findings advance our understanding of the neural mechanisms of our speech production system, which is capable of flexibly adapting to both linguistic and social contexts

    MAAIG: Motion Analysis And Instruction Generation

    Many people engage in self-directed sports training at home but lack the real-time guidance of professional coaches, making them susceptible to injuries or the development of incorrect habits. In this paper, we propose a novel application framework called MAAIG(Motion Analysis And Instruction Generation). It can generate embedding vectors for each frame based on user-provided sports action videos. These embedding vectors are associated with the 3D skeleton of each frame and are further input into a pretrained T5 model. Ultimately, our model utilizes this information to generate specific sports instructions.     Integration of Social Media News Mining and Text Mining Techniques to Determine a Corporate’s Competitive Edge

    Market globalization have triggered much more severe challenges for corporates than ever before. Thus, how to survive in this highly fluctuating economic atmosphere is an attractive topic for corporate managers, especially when an economy goes into a severe recession. One of the most consensus conclusions is to highly integrate a corporate’s supply chain network, as it can facilitate knowledge circulation, reduce transportation cost, increase market share, and sustain customer loyalty. However, a corporate’s supply chain relations are unapparent and opaque. To solve such an obstacle, this study integrates text mining (TM) and social network analysis (SNA) techniques to exploit the latent relation among corporates from social media news. Sequentially, this study examines its impact on corporate operating performance forecasting. The empirical result shows that the proposed mechanism is a promising alternative for performance forecasting. Public authorities and decision makers can thus consider the potential implications when forming a future policy

    Effects of phorbol myristate acetate and sivelestat on the lung injury caused by fat embolism in isolated lungs

    <p>Abstract</p> <p>Background</p> <p>Fat embolism syndrome (FES) associated with acute lung injury (ALI) is a clinical condition following long bone fracture. We have reported 14 victims due to ALI with FES. Our laboratory has developed an animal model that produced fat emboli (FE). The major purpose of this study was to test whether neutrophil activation with phorbol myristate acetate (PMA) and inhibition with sivelestat (SVT) exert protection on the lung.</p> <p>Methods</p> <p>The lungs of Sprague-Dawley rats were isolated and perfused. FE was produced by addition of corn oil micelles into the lung perfusate. PMA and SVT were given simultaneously with FE. Parameters such as lung weight/body weight ratio, LW gain, exhaled nitric oxide (NO), protein concentration in bronchoalveolar lavage relating to ALI were measured. The neutrophil elastase (NE), myeloperoxidase, malondialdehyde and phopholipase A<sub>2 </sub>activity were determined. We also measured the nitrate/nitrite, methyl guanidine (MG), and cytokines. Pulmonary arterial pressure and microvascular permeability were assessed. Lung pathology was examined and scored. The inducible and endothelial NO synthase (iNOS and eNOS) were detected.</p> <p>Results</p> <p>FE caused ALI and increased biochemical factors. The challenge also resulted in pulmonary hypertension and increased microvascular permeability. The NE appeared to be the first to reach its peak at 1 hr, followed by other factors. Coadministration with PMA exacerbated the FE-induced changes, while SVT attenuated the effects of FE.</p> <p>Conclusions</p> <p>The FE-induced lung changes were enhanced by PMA, while SVT had the opposite effect. Sivelestat, a neutrophil inhibitor may be a therapeutic choice for patients with acute respiratory distress syndrome (ARDS) following fat embolism.</p

    Maintaining CD4/CD8 ratio and Th1-CTL subsets of chimeric antigen receptor (CAR)-T cells in serum-free culture conditions

    Chimeric antigen receptor (CAR) T cells therapy is a promising strategy that significantly controlled the progress of cancer diseases. CAR-T cells could kill cancer cells through cellular immune response; therefore, CD8+ cytotoxic T cells are critical for CAR-T cell therapy. However, recent papers reported that CD4+ T helper cells were important for the response and maintenance of CAR-T cells in vivo. Here, we developed a serum-free CAR-T cell preparation process that maintained the T cell population and controlled the T cell subsets. The CD4+ and CD8+ T cell population in CAR-T cells were maintained at averagely 59.4 % and 34.6%, and the major T cell subsets were Th1 cells and cytotoxic T lymphocytes (CTLs), implying the potentially high cellular immune response. To verifying whether the prepared CAR-T cells were exhausted, the expression of several immune checkpoint markers was determined. Of interest, only less than 20% of CAR-T cells at endpoint were PD-1+ or CTLA4+, but more than 40% of CAR-T cells at the endpoint were TIM-3+, implying most CAR-T cells were not exhausted. These CAR-T cells produced more than 1 ng/mL of IFN-γ in the response to the antigen. Altogether, CAR-T cells could be prepared in our serum-free process in the controlling of T cell subsets, leading to potential high therapeutic potency.     Ring Chromosome 7 Presenting with Intrauterine Growth Restriction and Multiple Anomalies

    SummaryObjectiveRing chromosome 7 is a very rare chromosomal anomaly that may have a grave prognosis. Nevertheless, the clinical features associated with ring chromosome 7 are highly variable. Here, we report a case with ring chromosome 7 and the perinatal findings.Case ReportA 32-year-old, gravida 1, para 0, woman was referred to our hospital because of intrauterine growth restriction (IUGR) and oligohydramnios at 35 weeks of gestation. Prenatal ultrasound revealed a severe IUGR fetus presenting with multicystic kidney, hydronephrosis and oligohydramnios. At parturition, the birth weight of this male infant was 1,720 g, and a battery of anomalies were also noted, including imperforate anus, hypospadia, micropenis, right cryptorchidism, severe IUGR, multiple nevi on the forehead, shoulder and left thigh, brain atrophy, right multicystic kidney, and left mild hydronephrosis. Cytogenetic study from cord blood revealed a ring chromosome 7.ConclusionRing chromosome 7 is extremely rare and our case might be the 15th and youngest case in the medical literature. Our case had multicystic kidney and imperforate anus, which have not been reported previously. Prenatal diagnosis of ring chromosome 7 is very difficult. When fetuses present with severe IUGR, oligohydramnios and multicystic kidney, chromosomal aberrations should be kept in mind, and perinatal cytogenetic workup is warranted

    FACTORS AFFECTING THE TRANSFER OF INTER-ORGANIZATIONAL SYSTEMS TO CHINA: A CASE STUDY OF HIGH-TECH ENTERPRISES

    Due to the huge market potentiality, profuse production resource such as workforce and land, and low production costs in China, many international high-tech enterprises are moving their production line to China. Accompanying production offshoring is the transfer of Information Technology (IT) to ensure that the production processes remain efficient and effective at home and abroad. Many enterprises however encounter problems in transferring their IT to China, especially when the IT is an inter-organizational system (IOS), which is across organizational boundaries and involves the management of relationships among participants. In order to understand how to successfully transfer IOS to China, we aim to find out key relational factors of IOS performance in China through an observation of a binary relationship built upon an e-procurement system between a Taiwanese PC ODM (Original Design Manufacturer) and its Chinese suppliers. Based on the relational view of the firm, four relational factors are proposed and examined. The results show that IOS specific investments and effective IOS governance most strongly affects the result of IOS transfer to China, whereas complementary resource endowment has least influence

    In vitro high expansion of chimeric antigen receptor (CAR)-T cells in serum-free process conditions

    Manufacturing process is an important and complex factor for preparing chimeric antigen receptor (CAR) T cells for therapy. Although serum was widely applied in the culture or expansion of T cells, the quality of serum could be varied from batch to batch, leading to the variation of T cell expansion and quality. In addition, the safety of pathogens from serum and Chemistry, Manufacturing, and Control (CMC) were required to be considered. To overcome the disadvantages of serum application in T cell culture, serum-free and xeno-free culture conditions were required. We intended to develop a rapid serum-free culture condition for the expansion of immune T cells ex vivo. In our optimized serum-free condition, CAR-T cells could be expanded to about 100-200 times to the initial cell number after 6-day culture and the cell viability of all specimens was above 98%. Of interest, the percentage of CAR+ population in all specimens was increases, and the T cell pollutions could be maintained at averagely about 35-40% of CD8+ T cells and averagely about 50-55% of CD4+ T cells after culture. Taken together, our conditions could be applied in the expansion of CAR-T cells for cell therapy to support the minimum requirement of blood or cell samples from patients and to maintain the T cell population.
